hPSC colony

An hPSC (human pluripotent stem cell) colony is a cell aggregate where a cell does not exist on its own but rather consists of multiple cells.
Pluripotent stem cells such as iPS cells (induced pluripotent stem cells/iPSCs) and ES cells (embryonic stem cells/ESCs) self-renew while maintaining their undifferentiated state, and the cells adhere to each other and increase. Common cells proliferate from single cells to form colonies, but pluripotent stem cells vary depending on the culture method. In the conventional method, the grown colonies are not dissociated into single cells, but rather are crushed into a clump state of a population comprising about 50 to 100 cells when passaged. In recent years, depending on the culture method, passage may be performed by dissociating the cells into single cells. However, even when dissociating colonies into single cells, cells may gather to form a new colony. Therefore, when trying to perform cloning, it is necessary to take measures such as reducing the cell density or seeding a single cell in a well of multiwell plate.
